You're right about it being a two-syllable word - certainly no space in it to pronounce it the way they do in the series. I can also see it being pronounced with the accent on the first syllable as well (that's used for the plural form, usually).
I'd like to challenge your last sentence - there's no indication it's true (absence of evidence isn't evidence of absence), and it's also possible the extra trials simply hadn't been applied to anyone else.
It's a major point in Holt's career that he was an openly gay cop, and being an openly gay cop, he was chastised and constantly shunned because of that by his straight white male cop coworkers throughout his career. I don't think it's possible anyone in the precinct didn't know. See S1E16, where this is prominently stated
