In the Vampire Diaries S02E21, The Sun Also Rises, when Klaus's main werewolf turns, why doesn't Kyle himself and his backup werewolf also turn at the same time?

I think it's because werewolves turn on the full-moon night, not necessarily at the same time. The exact hour differs from person to person.

Proving something about a mythical entity may be a little tricky –  dav1dsm1th Feb 20 '14 at 12:40

well, I haven't seen that episode, hmmm ..vampire diaries to be specific! But talking about werewolf transformation in general, Don't in movies the werewolves transform usually faster when the moonlight shines directly/more on them (High exposure) or only when that happens (like in Vanhelsing) ?

And they always try to runaway from the direct moonlight while transforming as though trying to stop the transformation to happen quickly. The impact of moonlight is considered very much.

In Italy, France and Germany, it was said that a man or woman could turn into a werewolf if he or she, on a certain Wednesday or Friday, slept outside on a summer night with the full moon shining directly on his face.
